    Surveillance and evidence of contamination in hospital environment from meticillin and vancomycin-resistant microbial agents

    Introduction. Direct contact is undoubtedly the main means of transmission of hospital infections. An investigative study was therefore conducted to assess workplace surfaces at risk from microbial contamination. Methods. The study was conducted using swabs and contact slides placed on the palms of healthcare workers during their routine patient care and on workplace surfaces (e.g. telephones, computers, medication trolleys, taps) in treatment rooms, oper- ating theatres and wards. Disposable swabs were used for rapid screening and read with a bioluminometer. At the same time, a sample was taken from those testing positive using a contact slide. The samples testing positive for Staphylococci underwent identification to assess resistance to meticillin-resistant Sta- phylococcus aureus (MRS/MRSA) and to vancomycin (VISA/ VRSA) Results. Meticillin-resistant Staphylococcus strains were found on 14.7% (20/136) of samples taken from the hands of workers and 35.7% (15/42) of those from hospital surfaces. An even higher resistance to meticillin and/or vancomycin than that found for S. aureus was identified in nosocomial strains of coagulase negative staphylococci, including S. epidermidis and S. haemolyticus. Conclusion. The study concludes that there is thus a need for greater care in complying with procedures designed and support for surveillance to reduce the risk of infection

    Human papillomavirus vaccination coverage among adolescents living in southern Italy

    Objective: The aim of this study was to estimate HPV vaccination coverage in the target population residing in Sicily, five years after launch of the vaccination campaign, and to analyze its organization in this region. Methods: Regional data as at 31 December 2013, grouped by province, issued by the Regional Health Authority were used. The organization and information materials of the campaign were also assessed (letters, posters, brochures, etc.). Results: The results for Sicily show uptake rates for three doses of HPV vaccination of 56.5%, 55.8%, 58.2%, 55.3% for cohorts born in 1997, 1998, 1999, 2000 respectively, and 56.4% for cohorts born in 1996. These figures highlight the problems encountered during the promotion campaign and vaccination provision. Conclusions: Vaccine uptake in Sicily was lower than national figures for Italy as a whole for all cohorts and both fall far short of the targets set by the National Immunization Prevention Plan 2012-2014. In order to promote vaccination uptake and improve coverage, at both regional and local level, the quality of information should be improved and more communication campaigns be instigated to increase the involvement of professionals

    Hepatitis E virus (HEV) in sewage from treatment plants of Messina University Hospital and of Messina city council

    Samples of sewage from treatment plants at the ?G. Martino? University Hospital of Messina (AOU) and that of Messina City Council were analysed to detect the hepatits E virus. Samples were taken on sewage entering and exiting the treat- ment plants on a monthly basis over a one-year period from both the hospital plant (24 samples) and the municipal plant (22 samples). All sewage samples were pretreated by ultrafiltraton and concen- tration and finally processed by the PCR method to amplify gene material. A total of three samples tested positive: two (8.33%) entering the AOU treatment plant and one (4.5%) entering the municipal plant while no cases of HEV were detected in samples of treated sewage. These findings confirm the presence of the virus in the city of Messina and showed that the two treatment plants to be working efficiently when tested

    The puzzling properties of the MACS1149-JD1 galaxy at z=9.11

    We analyze new JWST NIRCam and NIRSpec data on the redshift 9.11 galaxy MACS1149-JD1. Our NIRCam imaging data reveal that JD1 comprises three spatially distinct components. Our spectroscopic data indicate that JD1 appears dust-free but is already enriched, 12+log(O/H)=7.900.05+0.0412 + \log {\rm (O/H) } = 7.90^{+0.04}_{-0.05}. We also find that the Carbon and Neon abundances in JD1 are below the solar abundance ratio. Particularly the Carbon under-abundance is suggestive of recent star formation where Type~II supernovae have already enriched the ISM in Oxygen but intermediate mass stars have not yet enriched the ISM in Carbon. A recent burst of star formation is also revealed by the star formation history derived from NIRCam photometry. Our data do not reveal the presence of a significant amount of old populations, resulting in a factor of 7×\sim7\times smaller stellar mass than previous estimates. Thus, our data support the view that JD1 is a young galaxy.Comment: Accepted for publicatio

    Interactome-Seq: A Protocol for Domainome Library Construction, Validation and Selection by Phage Display and Next Generation Sequencing

    Folding reporters are proteins with easily identifiable phenotypes, such as antibiotic resistance, whose folding and function is compromised when fused to poorly folding proteins or random open reading frames. We have developed a strategy where, by using TEM-1 \u3b2-lactamase (the enzyme conferring ampicillin resistance) on a genomic scale, we can select collections of correctly folded protein domains from the coding portion of the DNA of any intronless genome. The protein fragments obtained by this approach, the so called "domainome", will be well expressed and soluble, making them suitable for structural/functional studies. By cloning and displaying the "domainome" directly in a phage display system, we have showed that it is possible to select specific protein domains with the desired binding properties (e.g., to other proteins or to antibodies), thus providing essential experimental information for gene annotation or antigen identification. The identification of the most enriched clones in a selected polyclonal population can be achieved by using novel next-generation sequencing technologies (NGS). For these reasons, we introduce deep sequencing analysis of the library itself and the selection outputs to provide complete information on diversity, abundance and precise mapping of each of the selected fragment.     Eliminating congenital rubella: a seroepidemiological study on women of childbearing age and MMR vaccine coverage in newborns

    Introduction. Rubella can have particularly serious effects on the product of conception if contracted during pregnancy. Thus, the main aim of rubella vaccination programmes is to prevent infection during pregnancy. Materials and methods. A seroepidemiological study was conducted from July 2006 to December 2007 on 1,000 women of childbearing age, 15 to 45 years old, using specific rubivirus antibody assays, IgG and IgM. A questionnaire administered at the same time allowed us to survey how much women knew about this disease. In addition, MMR vaccine coverage rates were analysed for cohorts born in the local health districts of Messina for the period 1993-2006. Results. An analysis of the replies given to the questionnaire showed an estimated 42.8% of the women to have immunity from rubella, while the serological study showed an immunity coverage rate of 80.6%. Vaccination coverage in the local health districts regarding the first dose of MMR was 81% (cohorts 1993-2005), while the rate was only 24% for the second dose (cohorts 1993-2002). Conclusions. Both immunity coverage in women of childbearing age and that for newborns (for the cohort considered) fall below the 95% target set by the National Elimination Plan for Measles and Congenital Rubella (PNEM). It is therefore necessary to provide women with adequate information about the risks of rubella during pregnancy and about the benefits of vaccination, as well as to recoup subjects at risk or those whose immune status is unknown. Public health authorities also need to make continued efforts to increase the number of MMR vaccinations throughout the region

    The Grism Lens-Amplified Survey from Space (GLASS) X. Sub-kpc resolution gas-phase metallicity maps at cosmic noon behind the Hubble Frontier Fields cluster MACS1149.6+2223

    (Abridged) We combine deep HST grism spectroscopy with a new Bayesian method to derive maps of gas-phase metallicity, nebular dust extinction, and star-formation rate for 10 star-forming galaxies at high redshift (1.2<z<2.31.2<z<2.3). Exploiting lensing magnification by the foreground cluster MACS1149.6+2223, we reach sub-kpc spatial resolution and push the stellar mass limit associated with such high-z spatially resolved measurements below 108M10^8M_\odot for the first time. Our maps exhibit diverse morphologies, indicative of various effects such as efficient radial mixing from tidal torques, rapid accretion of low-metallicity gas, etc., which can affect the gas and metallicity distributions in individual galaxies. Based upon an exhaustive sample of all existing sub-kpc metallicity gradients at high-z, we find that predictions given by analytical chemical evolution models assuming a relatively extended star-formation profile in the early disk formation phase can explain the majority of observed gradients, without involving galactic feedback or radial outflows. We observe a tentative correlation between stellar mass and metallicity gradient, consistent with the downsizing galaxy formation picture that more massive galaxies are more evolved into a later phase of disk growth, where they experience more coherent mass assembly at all radii and thus show shallower metallicity gradients. In addition, we compile a sample of homogeneously cross-calibrated integrated metallicity measurements spanning three orders of magnitude in stellar mass at z1.8z\sim1.8. We use this sample to study the mass-metallicity relation (MZR) and test the fundamental metallicity relation (FMR). The slope of the observed MZR can rule out the momentum-driven wind model at 3-σ\sigma confidence level. We find no significant offset with respect to the FMR, taking into account the intrinsic scatter and measurement uncertainties.Comment: 26 pages, 14 figures, and 6 table

    CLASH: Weak-Lensing Shear-and-Magnification Analysis of 20 Galaxy Clusters

    We present a joint shear-and-magnification weak-lensing analysis of a sample of 16 X-ray-regular and 4 high-magnification galaxy clusters at 0.19<z<0.69 selected from the Cluster Lensing And Supernova survey with Hubble (CLASH). Our analysis uses wide-field multi-color imaging, taken primarily with Suprime-Cam on the Subaru Telescope. From a stacked shear-only analysis of the X-ray-selected subsample, we detect the ensemble-averaged lensing signal with a total signal-to-noise ratio of ~25 in the radial range of 200 to 3500kpc/h. The stacked tangential-shear signal is well described by a family of standard density profiles predicted for dark-matter-dominated halos in gravitational equilibrium, namely the Navarro-Frenk-White (NFW), truncated variants of NFW, and Einasto models. For the NFW model, we measure a mean concentration of c200c=4.010.32+0.35c_{200c}=4.01^{+0.35}_{-0.32} at M200c=1.340.09+0.101015MM_{200c}=1.34^{+0.10}_{-0.09} 10^{15}M_{\odot}. We show this is in excellent agreement with Lambda cold-dark-matter (LCDM) predictions when the CLASH X-ray selection function and projection effects are taken into account. The best-fit Einasto shape parameter is αE=0.1910.068+0.071\alpha_E=0.191^{+0.071}_{-0.068}, which is consistent with the NFW-equivalent Einasto parameter of 0.18\sim 0.18. We reconstruct projected mass density profiles of all CLASH clusters from a joint likelihood analysis of shear-and-magnification data, and measure cluster masses at several characteristic radii. We also derive an ensemble-averaged total projected mass profile of the X-ray-selected subsample by stacking their individual mass profiles. The stacked total mass profile, constrained by the shear+magnification data, is shown to be consistent with our shear-based halo-model predictions including the effects of surrounding large-scale structure as a two-halo term, establishing further consistency in the context of the LCDM model.Comment: Accepted by ApJ on 11 August 2014.     Systematic search for lensed X-ray sources in the CLASH fields

    We search for unresolved X-ray emission from lensed sources in the FOV of 11 CLASH clusters with Chandra data. We consider the solid angle in the lens plane corresponding to a magnification μ>1.5\mu>1.5, that amounts to a total of ~100 arcmin2^2. Our main goal is to assess the efficiency of massive clusters as cosmic telescopes to explore the faint end of X-ray extragalactic source population. We search for X-ray emission from strongly lensed sources identified in the optical, and perform an untargeted detection of lensed X-ray sources. We detect X-ray emission only in 9 out of 849 lensed/background optical sources. The stacked emission of the sources without detection does not reveal any signal in any band. Based on the untargeted detection, we find 66 additional X-ray sources that are consistent with being lensed sources. After accounting for completeness and sky coverage, we measure for the first time the soft- and hard-band number counts of lensed X-ray sources. The results are consistent with current modelization of the AGN population distribution. The distribution of de-lensed fluxes of the sources identified in moderately deep CLASH fields reaches a flux limit of ~101610^{-16} and ~101510^{-15} erg/s/cm2^{2} in the soft and hard bands, respectively. We conclude that, in order to match the depth of the CDFS exploiting massive clusters as cosmic telescopes, the required number of cluster fields is about two orders of magnitude larger than that offered by the 20 years Chandra archive. A significant step forward will be made when future X-ray facilities, with ~1' angular resolution and large effective area, will allow the serendipitous discovery of rare, strongly lensed high-zz X-ray sources, enabling the study of faint AGN activity in early Universe and the measurement of gravitational time delays in the X-ray variability of multiply imaged AGN.Comment: Accepted for publication in A&
